Denosumab is what I've been hoping will be available when I'm done with my forteo treatments. I'm almost done with my 2 years, although I have heard talk of forteo being extended to 3 years. That might tide me over muntil the denosumab will be on the market, it may be already for all I know. I'll ask my Dr. It sounds very promising as like the forteo (teriparatide) it is not a bisphosphonate. They actually build bone, although in different ways. My rheumatologist talked to me about some time ago but it was still in trial studies. It's being developed by Amgen, so you can check out their site or I also checked the New England Journal of Medicine. ( bunch of different Dr.'s research studies)

It is something called a biologic and is marketed by Amgen, it promotes bone growth rather than stopping bone loss, as I understand it. I've been watching it pretty closely since I just got diagnosed in April.

I knew it was a biologic and my son (genetics) explained it to me but I just looked it up again. A biologic is a drug made from genetic material like forteo, antibodies & proteins like denosumab, interleukins(don't know what they are & Dr. Son is not here) and vaccines. So I guess anything not made from chemicals like most drugs.

I've been following d-mab for about a year. It seems to work more like a bisfosphonate than terraparitide (forteo). I'll be finished with 2 years of forteo in November and I 'll need something to take. My endo suggested Actonel. Perhaps it will br d-mab. I wish I could take forteo for at least another year.

I think that it's more likely that this is acting more like a natural immune reaction against the osteoclast cells that destroy bone. This is an antibody, so I suppose it should attach to these bone killing cells and kill them in turn. Not at all like a bisphosphonate, really. Probably something you'd want to keep away from growing children though, unless you want a bunch of elephant men running around. That's my son's words. He's in for the 4th.

So far clinicals have included 11000 women, hope there was not serious side effects. But for me nothing is worse than Bisphosphonates, for me those meds were worse than Coumadin.

My over educated mouthy (but love-able) son has declared bisphosphonates intended toxins. I've never taken them, I was severe enough when diagnosed to be put on forteo. I had good luck with it and no side effects to speak of. At least not like the horror stories I've heard about the bisphos. I'm hopeful about the denosumab. Also like to be able to take the forteo for another year. I got 5 more months left of my 2 years.

Hey debbie59, last week my internist was trying to convince me to start on Forteo. I declined, as being on Coumadin a daily injection would amount to 365 bruises. I know he is concerned but I have never had a fracture and bruising is a side effect of Coumadin. I informed him about Donesumab and give him a print out, when it is approved I will be back. He did not know about Donesumab, nor did I expect him to have Donesumab information. I happily told him all about it. Two sub-q injections per year amount to two bruises, I can handle that, hopeful.

I bruise very easily too. In fact I'm having one of those mini facelifts the 16th and I'm totally expecting to be hiding for at least twice the normal time. (LOL) What we won't do!! I've not had that much bruising with forteo. It's a tiny tiny needle.

I thought after you take Forteo for 2 yrs , you have to start taking a Bisphosphonates after, as to seal in the good that Forteo did for your bones!! Or you will lose what Forteo has done!
This is all so confusing.

I took a one monthly Boniva last year, and was sick for two weeks.
I hope I never have to take anything like that again.
I'm just trying alternatives, and hoping it will work!

Re. Denosumab; thanks for posting the info ' however I found the studies a bit overwhelming -- so can anyone summarize the side effects?
I'm currently sticking to my regime of 'no drugs' (except for mega dosing D3 and the necessary calcium etc.) and weight bearing exercise, vibrational platform and eating prunes daily... I've only been diagnosed a couple of months ago - but my bone loss is major.
I reel much stronger since I started my regime (thanks fo Susan Brown's book and site) and am diligent about the akaline balance way of eating.
Is anyone (who is against the drugs) thinking of this new drug- since it's 'biologic' in nature?
Sigh, it's so confusing - but at least there's more to choose from.

Hey betterbones, in August when I see my Hematologist I am going to discuss Donesumab and have him do some side effects research. I do know it will not interact with Coumadin. Also I do know I cannot take any toxins such as the Bisphosphonates in any form.

Denosumab is a biologic drug (not chemical). it is a monoclonal antibody designed to block the actions of osteclasts, which attack the osteoblasts. Osteoblasts build bone and osteoclasts break it down. When we are growing this is a necessary function for bone turnover, otherwise no osteoclasts would cause bone growing out of control, like the elephant man. With osteoporosis the last thing we need are those buggers breaking down bone we've just made by taking Forteo for 2 years. So a lot of us are looking forward to the marketing of this new drug. Char 42 informed me that it should be on the market by October 09 or sooner. That's good news to me.
